Standard Operating Procedure: ABO Blood Grouping
1. Purpose: Determine patient ABO blood type for safe transfusion.
2. Specimen: EDTA whole blood (forward) and serum/plasma (reverse).
3. Reagents: Monoclonal Anti-A (blue), Anti-B (yellow), Anti-A,B; 2-5% A1 and B reagent red cells.
4. Procedure:
- Forward Typing: Add 1 drop patient 3-5% RBC suspension to tubes/slides with Anti-A and Anti-B. Mix, centrifuge (if tube), and read for agglutination.
- Reverse Typing: Add 1 drop patient serum to tubes/slides with A1 cells and B cells. Mix, incubate 5 mins (RT), centrifuge, and read.
